Reasons why PCR doesn't work?
« on: May 02, 2009, 02:11:14 AM »
My entire lab ran PCR, and none of the students' PCR came out.  We used a marker, positive control with lambda DNA, negative control with water, and our DNA sample.  Typically, the positive control and DNA sample would turn out illuminated, but after running the PCR, nothing came out.  Does anyone have any ideas on what may have went wrong?  Thanks in advance!

Many things could have gone wrong, but it's hard to tell from your description.  Since there's little you can do about it now, you're going to have to work out a description of what went wrong for the class write-up, I'm guessing.  So ... what do you know, about PCR, and how it works?  What do you need to perform a PCR, what are those things called, and what conditions are needed for the PCR reaction to run?
Hey, I'm not judging.  I just like to shoot straight.  I'm a man of science.

Could be helpful if you do the protocol and paste it here. Then we may analyze, what could go wrong..
