Boc deprotection..selectively deprotection?
« on: May 26, 2011, 05:00:36 PM »
Hey,

I am willing to carry out a Boc deprotection in my peptide, which has the follow sequence:

Fmoc-Lys(Boc)-Ser(tBu)-Ala-Gly-Ala-Wang Resin

Usually to deprotect Boc I employ TFA/TIPS, but then I will also cleave the Resin...and it's something that I don't want it...I will only cleave the Boc group in the Lysine, and not tBu as well as Fmoc and the resin...so I want to get....:

Fmoc-Lys-Ser(tBu)-Ala-Gly-Ala-Wang Resin

Any suggestion? lower % of TFA?

Thanks!

For N-Boc deprotection, you might be able to get away with TBAF1. I have no experience with the solid-phase synthesis but I guess the resin should be fine.
