Ammonium sulfate with potassium phophate
« on: August 29, 2013, 12:09:14 PM »
I'm making media with 10.4% potassium phosphate dibasic, 5.4% potassium phosphate monobasic and 2% ammonium sulfate. I get a heavy precipitate. The recipe is taken from a publication so I'm assuming I should be able to get full solubility. Is there an sequence order to the solubilization that will prevent precipitation?
Thanks.

Does a precipitate form when you mix the stock solutions?

Can you provide the recipe that you used (weights and volumes) from your lab book, not the publication?

Oh, dear. Now I feel stupid. I was using Ferrous Ammonium Sulfate. I did not see the ferrous. I repeated with Ammonium Sulfate and its not problem! Lesson learnt - make sure you are seeing the whole label!
Thanks for responding. It prompted me to check the label.

Don't feel stupid, I made a buffer for gel electrophoresis but didn't read that the solution I had made was a 100 x stock. Turned on the power, lots of bubbles and the gel melted :) we all make mistakes, stupid people don't learn from them.

All the best with your experiment.

Thank you! Its been over 10 years since my PhD and there seems to be an infinite number of mistakes to make. As you say, it keeps us learning.  :)
