thiamine biosynthesis
« on: July 20, 2015, 08:07:39 PM »
Dear member
Please educate me on this:
Is it possible to synthesize thiamine in an enzymatic reaction of hydroxymethylpyrimidine and thiazole through removal of hydroxyl group from hydroxymethylpyrimidine?
for structures please refer 6th page (2416) of this article: http://www.jbc.org/content/235/8/2411.full.pdf
If it is not possible; why?
Thanks!
With regards
Pari

Maybe a good place to start is to examine the chemical logic of the biosynthesis.  Why do you suppose nature chose to use pyrophosphate as the leaving group?  What chemical advantages do you see?
