ratio of homogenization buffer?
« on: August 03, 2016, 12:43:30 PM »
I have a very basic question that is driving me crazy. I´ll need to run some enzyme assays, and I´m getting stuck in the first step.

I´m going to homogenize some tissue, specifically samples that weight 50mg. The protocol says ..."homogenize in 1:5(w/v) of the homogenization buffer".

My guess here is, being 1:5 w/v= 20% w/v, which means 20g tissue per 100ml of homogenization buffer.

Is it ok so far? Then, transformed into my amount of tissue it´d be: 20gtissue/100mlbuffer=0,05gtissue/x...0,250ml homogenization buffer!

Am I wrong here?
Thanks in advance! ;)

I don't see any errors.  The 1:5 ratio is similar to what we use for sonnication of bacteria.
