Activated Charcoal: Better to use in water or xylene?

Im looking to purify my solution and these seem to be my options:

Either i can add the charcoal to the alkaloid saturated xylene, or i can wait till i salt the alkaloids into water and add it then. Which is the better medium to minimize alkaloid loss and maximize purification?

Also i was told that to get the most out of a charcoal filtering, you have to leave it in your solution in a hot water bath overnight. Is that true? Do i risk unncessary alkaloid loss by doing so?

No one knows the answer to this? I could really use one as i have this step coming up within a few hours and have to know the most optimal route.

Add that in aqueous. The adsorption is better in aqueous and ~40 Deg C
