Help- Boc/peptide solution gets yellow...

Hi folks,

i wan't to BOCylate peptide and i don't know if this is good or not (never had this phenomenon):

i had ~ 50µg Peptides disolved in 25µl DMSO.. then i added 100µl BOC (disolved in DMSO, high concentration) and 6,25µl DIPEA as Base...

Now the solution (already incubated for 5 hours) is pale yellow/brown ( didn't add anything...)

is that normal ? or what could be the reason for it ?

Hard to say.  I wouldn't say its not normal, as a reaction probably occurred or there exist a  chromophore that has been activated with BOC protection.  But the color can be the result of many things (bad/old reagents DMSO, DIPEA, DIBOC, peptide).  As long as you can exclude contamination, then all you need to do is characterize your product.

ok thx, i think it's due to the DIPEA.. but well i'll analyze it...
