Topic: Need help finding concentrations of protein, dye and complex in solution  (Read 3453 times)

A Solution contains 1.00*10^-4 M total protein
2.00*10-3M total dye

Absorbance at 280nm = 1.4
L = 1cm

Protein + dye = complex

Molar Absorption (E):
E(Protein) = 12000/Mcm
E(Dye) = 0 /Mcm
E(complex) =15000 /Mcm

I know the beer-lamber law

A=ECL

But how do I use this equation to find the concentration of protein,dye, and complex in solution.

Try to devise formula for total absorbance as a function of concentrations of the protein, dye and complex. Concentrations of the dye and coplex and of the proteing and complex are related - how? These will give you set of equations that you have to solve.
