compound seperation
« on: August 11, 2010, 03:49:15 AM »
Hello
I am having problem with the compound seperation. I am working with the hexane extract of the cudrania tricuspidata. But i coluld not seperate a single compound. I always get two compounds having same RF value. Its been quite long time that I am trying to seperate the compounds. I tried all possible solvents like toulene, hexane,ethylacetate,etc...but I colud not seperate. If anybody has idea about seperating the compounds that co-spot in TLC please help me.
Any type of suggestion will be appriciated.
Thank you friends.

Did you try a dichloromethane:hexane mixture?

Thank you for the reply. I already tried this solvent system but could not get the satisfactory seperation. If you have any idea about seperating thise kind of non polar compounds then please suggest me.
Thank you very much.

Have you tried normal phase?

I cannot use reverse phass because my compound doesn't move in RP colum at all. SO I always have to use normal phase silica for the seperation. If you have any idea then please suggest me.
Thank you very much.

Have you tried using a different solid phase e.g. alumina in place of silica?

Try and separate therm on a prep HPLC system, at the least you can get a baseline separation which you may be able to apply to a column
Development Chemists do it on Scale, Research Chemists just do it!

I always get two compounds having same RF value.

Do you have any idea of what these compounds are or what functional groups are present? You may be able to derivatise one (or both) component(s) of the mixture to get seperable compounds.

Crystallisation maybe?

Thank you for your reply. I have no idea about these compounds. They are oily liquid. These are soluble in hexane or chloroform. If you have any idea about the crystallization of these compounds than please suggest me.
Thank you so much.

Do you have NMR or IR data of the mixture? It probably helps you to predict the functional groups or structures of both. Do derivatisation first, follow by separation may be better.

If you know the possible functional group present in the compound of your intrest(of course in your case it is mixture!)
You can try for acid base extraction(provided that the compound (i)stable in acid and base conditions (ii)availability of sufficient amount of sample).
