Method for isolation of Chlorophyll
« on: June 23, 2011, 09:48:08 AM »
My question is if the following method is good for isolaiting clorophyll without damaging his internal structure (and wthout removing the Mg2+ from it):
1)Crushing spinach leaves in a 96%Ethanol solution.
2)Filter the solution to remove solid residues
3)Put the solution into a small matrass and make the pressure decrease with an external pump
4)Heat the matrass about 40-50ÂșC until all ethanol evaporates while the pump is working.
*The points 3-4 are something like using a "rotavapor" so that i wouldn't have to heat too much in order to don't change clorophill structure.

Is this a good method?Would you change something? (or if there's a better one pleas tell me, but I have to do that
without using strange lab tools)

Its not a bad way to start. How pure do you need your chlorophyll? Using this method you will get a crude extract that also contains many other chemicals from the plant (carotenoids, etc...)
Individual results may vary

Hi,
There's no problem on containing carotenoids, xanthofills etc. In fact the only condition is that the extract doesn't contain anything poisonous for a plant.
Ah, wich temperature do you think that is the maximum that chlorophyll can support?
Thank you
PS: I have just ended my first organic chemistry subject, so I don't know many things about organic chemistry.
