Help me to messure the melting curve of nucleic acid analogues

I want to messure the melting curve of nucleic acid analogues containing uracil and adenine as base. Can you tell me what kinds of buffer solution I should use. Does anyone have experience of observation of the formation of base pairs by NMR. Thanks in advance!

Why are you using NMR? Can't you just look at the change in absorbance at 260nm?

I can not see the change of absorbance in UV when I use water as solvent. I hope to see the chemical shift of proton in N-H in the base pairs moves in NMR.
