72% sulfuric acid hydrolysis for leaves
« on: July 09, 2014, 11:36:57 AM »
Hello,
I have a question regarding the sulfuric acid extraction for measuring lignin content.
Usually people hydrolyze plant tissue using 72% sulfuric acid to get the lignin content but they extract the tissue first and remove undesirable compounds using benzyl alcohol or acetone or some alcoholic solvent.

I wonder if I can skip the first extraction procedure and do the sulfuric acid hydrolysis to remove phenolic compounds, tannins, waxes, and cutins. Can anyone tell me among above compounds, which compounds 72% sulfuric acid can solublize during the hydrolysis.

Thanks!
