I don't understand the calculation of atomic mass unit.To calculate atomic mass unit,we use (1/12)x((12)/(6.02x10623)).why we need to use mass of an atom carbon-12 divide with the Avogadro constant.

I think you have mistyped that equation.

The atomic mass unit is defined as 1/12th of the mass of a carbon atom.

Mass of a carbon atom = 0.012/(6.02 x 1023) kg

(Or 12/(6.02 x 1023) g)

=> atomic mass unit = (1/12) x 0.012/(6.02 x 1023) kg

Or (1/12) x 12/(6.02 x 1023) g
