Hi,

I need to dfeprotect a primary TIPS alcohol off a nucleoside. I have tryed TBAF (which works), but it leavse a whole bunch of salts in the sample, and it si impossible to purify by column.

I want to try HF/Pyridine as a reagent. Does anyone know, if you can make it from the 48 % HF solution and pyridine, or does it have to be anhydrous?

Anhydrous HF is needed. HF/pyridine reagent is commercially available.

Thanks for your input. Just out of curiosity. What happens, if there is water present with HF-pyridine? Is the reagent ineffective for some reason, or does it damage other functional groups?

Anhydrous HF has completely different properties in comparison to its water solution
