Help me to messure the melting curve of nucleic acid analogues

I want to messure the melting curve of nucleic acid analogues containing uracil and adenine as base. Can you tell me what kinds of buffer solution I should use. Does anyone have experience of observation of the formation of base pairs by NMR. Thanks in advance! :)

Why are you using NMR?  Can't you just look at the change in absorbance at 260nm?

I can not see the change of absorbance in UV when I use water as solvent. I hope to see the chemical shift of proton in N-H in the base pairs moves in NMR.
