so, i have plotted my 1) initial velocity (v) against substrate concentration ,S.
2) Lineweaver-burk plot)
3)Eadie-Hofstee plot
4)Woolfe plot
And was asked to calculate the michaelis constant (Km) and the maximum velocity (Vmax) from each plot.
I understand what each graph demonstrates in terms of for eg :
1) Shows Vmax and Km ( This one i can do)
2) o y intercept = 1 / Vmax (This one, i dont know how to get Km and Vmax from all this information)
o gradient = Km / Vmax
o x intercept = -1/ Km
3)
o y intercept = Vmax (This one , i know the Vmax, and i think i know the Km, its just changing signs right?)
o gradient = -Km
o x intercept = Vmax / Km
4)
o y intercept = Km / Vmax (This one i know the Km value [its just the opposite sign right?] , but dont know gow to get Vmax)
o gradient = 1 / Vmax
o x intercept = -Km
If you could help me in the areas i have doubts, it would be appreciated.
