Hi guys,

We did HPLC of caffeine, adding pH=3 phosphate buffer. and we chose wavelength=254nm

And i have a question from lab report:

Discuss how the choice of the monitored wavelength affects the sensitivity of the analysis. Would you choose the same wavelength to quantify caffeine and benzoic acid? Why or why not?

What if anything do you know about the UV absorbance of Caffeine and Benzoic acid?

I think I got it,
as absorbance increases sensitivity also increases.

And for second question, no need to change wavelength, because in hplc substances are already separated and no imposition of absorbances will occur.
