Hello,

I feel like I am offering some snow to them polar bears on the North Pole...

We have finished a project and currently have underutilized capability for preparative work. If you have one molecule too many to handle, we can *delete me*


For analytical and semi-prep work we primarily use reverse phase chromatography implemented on a gradient capable Gilson Semi-preparative HPLC System.
It accommodates columns with 2-20 mm ID with corresponding flow rates of 0.2-20 ml/min and loading capacity ranging from micrograms to tens of milligrams

For larger samples, up to several grams, we use a a Fast Centrifugal Partition Chromatography (FCPC) instrument manufactured by the French firm KROMATON.
FCPC is unique because no solid support is used for the stationary phase. Its advantages listed below enable us to isolate almost any compound that can be eluted in an analytical HPLC run.

Absence of the irreversible adsorption enables recovery of all components of a mixture intact. This feature is priceless when it comes to the bioactivity-directed isolation of unknown compounds as it prevents loss of activity from fractionation.

Efficient purification of labile compounds is afforded by temperature or pH control of compatible solvent system.

High capacity; the instrument can accept a sample of up to 5 grams with very modest solvent consumption.

Tunable selectivity. It can be optimized for a given purpose by composing a solvent system appropriately. In complex cases requiring multi-step purification it can greatly complement other chromatographic techniques and as a rule should be used as the first step purification. FCPC can often separate compounds that have the same retention times on the solid phase.

Applicable to a wide range of polarity of chemical compounds. From unsaturated hydrocarbons or lipids on one end of polarity to the highly hydroxylated compounds such as saponins, tannins, proanthocyanidins, or even quaternary alkaloids on the opposite end of polarity range.

Applicable to the water-sensitive compounds. Several non-water containing solvent systems have been described in literature.

Most of time we monitor on-line absorbance at 250 nm to direct fraction collection. Also use GC-MS and LC-MS for off-line and ID purity verification.
