1. The reaction catalyzed by RNA (or DNA) polymerases. The direction of the polymerization process (which
end is elongated?). The significance of hydrolysis of PPi released as one of the reaction products.


2. The interactions that stabilize DNA double helix – the role of hydrogen bonds and base-stacking interactions. The major structural parameters of DNA B-type helix (directions of strands, complementarity rules, number of base pairs (bp) per turn, grooves).


3. The reaction catalysed by an endonuclease. Which bond is hydrolysed? What are the products?


4. The reaction catalysed by a 5’to 3’ exonuclease and 3’to 5’ exonuclease.


5. Denaturation of DNA. Denaturing agents. Which interactions are broken? Is the process reversible?


6. Define the term: hyperchromic effect. What is the practical use of this effect?


7. What can we conclude from the sigmoidal shape of the DNA melting curve? Why AT-rich regions have lower melting temperature than CG-rich ones?

Also, the first two are basically complete statements.  We need you to actually ask us questions.  What is asked, what can you figure out, what can't you figure out, and we'll see if we can help.  Problem is, if all you want is a definition, all we can do is give it to you -- and you might as well check your book or class notes, it will be faster, because they're right in front of you.
